18-P003 A gene regulatory network for mesoderm directed by zebrafish No tail

چکیده
The secreted protein Hedgehog (Hh) acts a morphogen, forming a concentration gradient and controlling cell fate decisions in various developmental stages in many animals. In recent years, there has been significant interest in the role that glypicans play in Hh gradient formation and signalling; with various experimental studies demonstrating an effect on Hh transport, Hh stability and downstream transcription factor (CI) activity. Using mathematical modelling and analysis, we investigate the effects glypicans (e.g. Dally, Dlp) have on gradient formation and target gene expression domains. The incorporation of Hhglypican complexes yields models whose behaviour matches current experimental data more accurately than previous models. Furthermore, we show that interaction with glypicans enhances the robustness of

A gene regulatory network directed by zebrafish No tail accounts for its roles in mesoderm formation.
Using chromatin immunoprecipitation combined with genomic microarrays we have identified targets of No tail (Ntl), a zebrafish Brachyury ortholog that plays a central role in mesoderm formation. We show that Ntl regulates a downstream network of other transcription factors and identify an in vivo Ntl binding site that resembles the consensus T-box binding site (TBS) previously identified by in ...
متن کاملMolecular identification of spadetail: regulation of zebrafish trunk and tail mesoderm formation by T-box genes.
Inhibition of fibroblast growth factor (FGF) signaling prevents trunk and tail formation in Xenopus and zebrafish embryos. While the T-box transcription factor Brachyury (called No Tail in zebrafish) is a key mediator of FGF signaling in the notochord and tail, the pathways activated by FGF in non-notochordal trunk mesoderm have been uncertain. Previous studies have shown that the spadetail gen...
متن کاملIdentification of direct T-box target genes in the developing zebrafish mesoderm.
The zebrafish genes spadetail (spt) and no tail (ntl) encode T-box transcription factors that are important for early mesoderm development. Although much has been done to characterize these genes, the identity and location of target regulatory elements remain largely unknown. Here, we survey the genome for downstream target genes of the Spt and Ntl T-box transcription factors. We find evidence ...
متن کاملAn interacting network of T-box genes directs gene expression and fate in the zebrafish mesoderm.
T-box genes encode transcription factors that play critical roles in generating the vertebrate body plan. In many developmental fields, multiple T-box genes are expressed in overlapping domains, establishing broad regions in which different combinations of T-box genes are coexpressed. Here we demonstrate that three T-box genes expressed in the zebrafish mesoderm, no tail, spadetail, and tbx6, o...
متن کاملStructure of the zebrafish snail1 gene and its expression in wild-type, spadetail and no tail mutant embryos.
Mesoderm formation is critical for the establishment of the animal body plan and in Drosophila requires the snail gene. This report concerns the cloning and expression pattern of the structurally similar gene snail1 from zebrafish. In situ hybridization shows that the quantity of snail1 RNA increases at the margin of the blastoderm in cells that involute during gastrulation. As gastrulation beg...
